The Effect of Purified Carrageenans on the Phagocytic Function of RES in Rats

Abstract
Although carrageenan, sulfated polygalactans extracted from marine algae, is a potent suppressor of immune response, a limited information is available as to the effect of the purified carrageenan on phagocytic function of RES in vivo.
The purpose of this study is to determine the comparative effects of purified iota, lambda and kappa carrageenan on the phagocytic activity of RES in rats. After a single intraperitoneal injection (12.5 at/1008 of body weight) of each carrageenan, the evaluation of phagocytic activity with carbon clearance method and the histologic examination of RES organs were performed at various time intervals, starting at 10 hours and ending at the 4th week.
The results were obtained as follows.
1. After an injection of iota carrageenan, there was a transient reduction in the phagocytic activity in the first and the third day. However, by the end of the lst week, a complete recovery in the activity was observed.
2. In lambda carrageenan-treated rats, there was a transient increase in the phagocytic activity on the first day, subsequent to which there was a reduction in phagocytic activity. However, by the end of the second week, an almost complete recovery in the activity was achieved.
3. The kappa carrageenan showed the most potent suppressive activity. Ten hours after injection, there was a severe reduction in the phagocytic activity, which never fully recovered during the entire experimental period.
4. In histologic examinations of liver and spleen, focal necrosis of the liver was observed on the 1st day after carrageenan injection and the active phagocytosis of carrageenan by macrophages was observed on the third day. Also, marked reduction in the number of splenic lymphocytes was seen after the two weeks. The degree of these lesions was more prominent in kappa carrageenan-treated rats.
